He completed his PhD in Chemistry at the University of Manchester in Chemistry\u00a0in 2016 with a thesis entitled \u201cStructure-Guided Directed Evolution of Alkyltransferase Enzymes\u201d after four years in the Micklefield group. Matthew has continued as a postdoctoral researcher in the Micklefield group and his research is focussed on developing Methyltransferases for Biocatalytic purposes.<\/p>\n<p>Matthew has worked with a number of different methyltransferases including the Catechol-O-Methyltransferase (COMT) enzyme, improving the regioselectivity of the enzyme through mutagenic and oligomeric separation techniques (see \u201c<strong>Effects of Active Site Modification and Quaternary Structure on the Regioselectivity of Catechol-<em>O<\/em>-Methyltransferase.\u00a0<\/strong>B. J. C. Law,\u00a0<strong>M. R. Bennett<\/strong>, M. L. Thompson, C. Levy, S. A. Shepherd, D. Leys, and J. Micklefield\u00a0<em>Angew. Chem. Int. Ed.\u00a0<\/em><strong>2016<\/strong>, <em>55<\/em> (8), 2683. (<a href=\"http:\/\/dx.doi.org\/10.1002\/anie.201508287\">http:\/\/dx.doi.org\/10.1002\/anie.201508287<\/a>)\u201d), and focussing on key enzymes involved in the alkaloid biosynthetic pathway (Coclaurine-<em>N<\/em>-methyltransferase and tetrahydroprotoberberine <em>cis<\/em>&#8211;<em>N<\/em>-methyltransferase).<\/p>\n<p>Further to methylation, he has also used the methyltransferases to perform alkylation reactions working with both enzymatic and chemical methods to synthesise SAM analogues, which coupled with methyltransferases can perform alkylation reactions. Finally, Matthew has worked as co-first author with Dr Anna-Winona Struck on developing a cascade reaction with COMT and mushroom tyrosinase that has been used to methylate and alkylate tyrosine residues on both peptides and proteins (see \u201c<strong>An Enzyme Cascade for Selective Modification of Tyrosine Residues in Structurally Diverse Peptides and Proteins.\u00a0<\/strong>A.-W.
